The FISH protocol consists of 3 main procedures: cells/tissue preparation, probe hybridization, and washing (post-hybridization).
It is critical for the hybridization step to have all optimal conditions to have a successful in situ result, including elevated temperature and elongated time of the hybridization reaction (usually overnight).
These two factors increase the possibility of buffer evaporation during the process, leading to drying out of the sample.
